The Terriers will at least compete for a Premier League place via the play-offs, and could even steal an automatic promotion spot from Bournemouth if the Cherries slip up.
Barnsley, meanwhile, will join Derby in third tier, bottom of the Championship with 30 points from 42 games.
It marks a huge change in fortunes for both sides, with Huddersfield finishing two places from the relegation zone in 20th last season, while Barnsley finished fifth.
Expectations were low at the John Smith Stadium after last season’s relegation, but the club kept faith in Carlos Corberan and were rewarded.

Valerien Ismael had taken the team to an astonishing finish in the play-offs after nearly being relegated from the Championship the previous season, and he was later snapped up by West Brom.
His replacement, Markus Schopp, only lasted until November, while Ismael didn’t fare much better with the Baggies, who were sacked in February before joining Besiktas.
Two other managers coached the Tykes to finish the season, but neither was able to make a difference enough to ensure the team’s survival.
